Margarita Vanderpoel may have been born as early as 1647. She was the daughter of one-time Beverwyck and Albany residents Wynant G. and his Tryntje Melgerts Vanderpoel.

About 1681, she married Albany tradesman Johannes Van Zandt. Their eleven children were born between 1682 and 1703. In 1683, she was a member of the Albany Dutch church.

During the 1690s, her family relocated to New York City.

Margarita Vanderpoel Van Zandt was living in Newtown Queens in 1724 when she was left personal property and an annuity in her husband's will. Johannes Van Zandt died later in 1724. Family sources state that Margarita died in October 1724.
